"Ficken Sie mich" is grammatically correct, but it is rather formal way to say a rather informal thing. Using the verb with an "en" ending and the formal pronoun "Sie" (with a capital) is a polite way of speaking to someone who is much older/younger than you, your boss/employee, or some other professional-type relationship. "Fick mich" is the causal, normal everyday way to say it, that is more likely to be used any sexual context, unless the character is having sex with their CO, and you're looking for a comedic effect. (Kind of like saying "Fuck me sir"). If there is more than one partner, your telling to fuck you, you'd say "Fickt mich". And as a bonus, you can say "fick dich" to tell someone "fuck you", or again, if more than one person: "fickt euch" ("ficken Sie" if you wanted to use the formal version for some reason).
